Paroxetine

For an informed discussion with your doctor or pharmacist. This information collection is not a clinically validated interaction checker. Missing data do not establish safety. Do not change or stop prescribed medicines on the basis of this information. The findings apply to the specific conditions studied; laboratory and animal studies cannot be directly extrapolated to humans.

Active substance: Paroxetine

Examples of brand names: Seroxat [DE]

ATC code: N06AB05

Main use (for orientation): Depression; anxiety

Documented research summary

A severe hypotension case involved anaesthetics, cannabis, amphetamines and paroxetine at the same time; the paediatric reassessment rates causality for the pair as doubtful. Another intoxication case involved cannabis and at least five medicines. Neither establishes isolated paroxetine–cannabinoid pharmacokinetics. The 2004 panic-disorder cohort showed a similar response to paroxetine following cannabis-associated onset of illness, but did not reliably examine continued, defined co-exposure. Comparator arms and radiolabelled paroxetine as a SERT marker are not therapeutic drug–drug interactions.

Uncertainties and limitations

No measured cannabinoid-related increase in paroxetine concentrations identified.

PMC11668912 contains shifted reference numbers; the original title and DOI of the hypotension case were checked separately. DIPS=1 indicates uncertain causality, not an established mechanism.

No class-specific evidence of safety for rare cannabinoids.

Separately recorded findings

2 separately recorded findings. Each statement applies to its documented study context.

Cannabis; composition not quantified – insufficient data

Finding ID: B0423

Type of result: Multiple-substance context, no isolatable drug–drug interaction

Observation: Confusion, hallucinations and sinus tachycardia after cannabis and a powder containing several prescription medicines.

Mechanism: Combined exposure; the mechanism of an individual pair cannot be determined

Study type: Multiple-substance intoxication case

Context studied: 16-year-old female patient; analysis identified several antidepressants and trihexyphenidyl in the powder.

Clinical interpretation: Interpret within the stated study context; no independent dosage or treatment recommendation.

Limitations and uncertainties: No isolated pair finding; individual quantities unknown and extensive exposure to multiple substances. Listed as safety context, not as an established cannabinoid interaction.

Cannabis/marijuana; composition unknown – insufficient data

Finding ID: B0425

Type of result: Uncertain case reassessed in a secondary source

Observation: Severe hypotension was described in a surgical setting; a later structured case assessment rated interaction causality as doubtful (DIPS=1).

Mechanism: Multiple exposures including anaesthetics and amphetamines

Study type: Anaesthesiology case report, with details obtained through a secondary review

Context studied: Paediatric case involving paroxetine, cannabis, amphetamines and perioperative medicines.

Clinical interpretation: Interpret within the stated study context; no independent dosage or treatment recommendation.

Limitations and uncertainties: Original abstract/full text unavailable; details explicitly obtained from Chapin 2025, not independently extracted from the original. Reference numbers in the secondary source are inconsistent; original title/DOI checked. No established pharmacokinetics for the pair.
