Bupropion

For an informed discussion with your doctor or pharmacist. This information collection is not a clinically validated interaction checker. Missing data do not establish safety. Do not change or stop prescribed medicines on the basis of this information. The findings apply to the specific conditions studied; laboratory and animal studies cannot be directly extrapolated to humans.

Active substance: Bupropion

Examples of brand names: Elontril [DE]; Zyban [DE]

ATC code: N06AX12

Main use (for orientation): Depression; smoking cessation

Documented research summary

FDA Epidiolex label CL02: CBD reduced bupropion AUC by 20% and Cmax by 19%, with hydroxybupropion unchanged. This contradicts a blanket claim that CBD increases every CYP substrate. A small withdrawal experiment showed worse mood with bupropion during cannabis withdrawal, with few effects during active smoking.

Uncertainties and limitations

The bupropion withdrawal effect is not identical to an acute pharmacokinetic interaction; the new urine-ratio paper without pair figures was not extrapolated.

Separately recorded findings

1 separately recorded finding. Each statement applies to its documented study context.

CBD – clinically studied

Finding ID: B0004

Type of result: Positive finding / indication

Observation: AUC −20%, Cmax −19%; hydroxybupropion unchanged.

Mechanism: CYP2B6; complex net effect

Study type: Human pharmacokinetics according to product information

Context studied: CBD 7.5 mg/kg twice daily; bupropion 150 mg; n not stated.

Clinical interpretation: Small in the regimen investigated

Limitations and uncertainties: Higher CBD exposure remains unresolved. EU prescribing information section 4.5 assesses the measured reduction as within the observed variability and not clinically significant. Do not infer general safety clearance.
